The 4416th was activated in June 1963 with the primary mission of testing new photo recon equipment, materials and tactics. The squadron initially performed its R&D missions on test beds RF-84 (2), RB-66 (2) and RF-101C. The squadron personal performed initial intallation and testing of the 1181 Mod to the RF-101, and tested the operational capabilities of the various new Recon equipment and systems (including films) for acceptance by the USAF. Two RF-4C aircraft were added at some date prior to 1966.

In addition, electronic recon equipment and tactics were evaluated on the RB-66. At the time the 4416th was activated under the 4411th CCTS, the fleet of RB-66s in the 4415th and 363rd were being stripped and prepared for mothballing.
